Research: Cytoskeleton-Membrane Interactions; Membrane Protein Structure and Function

Work in Anne Hitt's lab is targeted towards identification and characterization of proteins that connect the plasma membrane to the underlying cytoskeleton. Such interactions control many fundamental cellular processes, e.g. cell-cell adhesion and signal transduction. Her lab is focusing on the transmembrane protein ponticulin, the major high-affinity link between the actin-based cytoskeleton and the plasma membrane in Dictyostelium. She has cloned the gene for ponticulin from Dictyostelium and subsequently generated a cell line lacking ponticulin by homologous recombination. Currently the structure and biosynthesis of ponticulin are being investigated by site-directed mutagenesis.
